Resveratrol became famous as "the wine compound" in the early 2000s after research suggested it might explain the French Paradox. The actual science of resveratrol and skin aging is more interesting — and has almost nothing to do with wine. Here is what it actually does, and what dose it requires.

What Resveratrol Is

Resveratrol is a stilbene polyphenol produced by plants as a defense response to stress (fungal infection, UV exposure, injury). The highest natural concentrations are found in Japanese Knotweed (Reynoutria japonica), grape skins, and blueberries. It exists in two isomers: trans-resveratrol (bioactive) and cis-resveratrol (inactive). Only the trans form is biologically relevant for anti-aging purposes.

Why Wine Doesn't Work

A glass of red wine contains approximately 0.3-1.5mg of resveratrol. The doses used in longevity and anti-aging research range from 100mg to 1000mg of trans-resveratrol daily. You would need to drink 100-500 glasses of wine to reach therapeutic resveratrol doses — at which point the alcohol toxicity would far outweigh any resveratrol benefit. The "French Paradox" marketing was based on population observations, not mechanistic resveratrol biology. The wine connection is a correlation, not a causal mechanism.

The Actual Skin Mechanisms

1. SIRT1 Activation

Resveratrol's primary mechanism is activation of SIRT1 — the same sirtuin protein that NAD+ restoration supports. SIRT1 suppresses MMP-1 collagenase transcription, reduces oxidative stress, and regulates the inflammatory pathways that accelerate dermal aging. Trans-resveratrol at therapeutic doses measurably increases SIRT1 activity in human cells. This is synergistic with NAD+ supplementation: NAD+ provides the substrate SIRT1 needs, while resveratrol activates SIRT1 directly.

2. Glycation Inhibition

Resveratrol is one of the most studied natural inhibitors of advanced glycation end-product (AGE) formation. Glycation is the non-enzymatic bonding of glucose molecules to proteins — including collagen. When collagen becomes glycated, it cross-links into a stiffer, more brittle structure. AGEs also activate RAGE (receptor for AGEs), triggering inflammatory signaling that further degrades the matrix.

By inhibiting AGE formation, resveratrol protects newly synthesized collagen from the structural degradation that would otherwise occur as glucose molecules accumulate over time. This is particularly relevant in the integration phase of a 90-day collagen rebuilding protocol.

3. Antioxidant and UV Protection

Resveratrol has direct antioxidant activity and has been shown to modulate the oxidative stress response in UV-exposed fibroblasts. It does not replace sunscreen but provides complementary protection against the oxidative component of UV damage at the cellular level.

The Source Matters: Japanese Knotweed vs Grape-Derived

The most potent and consistent natural source of trans-resveratrol is Japanese Knotweed root extract, standardized to 98% or higher trans-resveratrol content. Grape skin extracts typically contain 20-50% trans-resveratrol at best. Standardization to 98% trans-resveratrol from Japanese Knotweed guarantees the bioactive isomer at consistent concentrations per dose — which grape-derived sources cannot reliably deliver.

Effective Doses

Human bioavailability studies place the effective dose range at 100-300mg trans-resveratrol daily for anti-aging mechanisms. Higher doses (500mg+) have been studied but show limited additional benefit for most outcomes.
